Hundreds of genes. Thousands of samples. One solution.™

Plexigen’s geneCube™ platform is ideal for gene expression studies involving a small to medium number of genes in a large number of samples. The platform has also been used for assaying a number of antibodies against a large number of antigens, equivalent to a parallel ELISA and for SNP genotyping if a small to medium number of SNPs and large number of samples are involved in an experiment.
Services are offered through the following two channels:

1. Direct service from Plexigen: A turn-key solution from experimental design to data analysis and a true customized service.

2. Service through Plexigen’s Marketing Collaborators: Plexigen is installing the geneCube platform at a number of qualified microarray service companies. Researchers can use the companies to process their samples.

Direct service from Plexigen

We can provide all of the advantages of our platform as a direct service to our customers. Currently we offer full service gene expression, genotyping and proteomic testing, including optional data analysis.

Gene Expression Experiments Using geneCube Platform

Plexigen's geneCube platform is infinitely flexible for experimental design, which can meet customer's needs in gene content and accuracy requirements. A geneCube is essentially a deck of cards with each card coated with one or a number of oligo probes. The deck can be formed according to customer's gene content needs. Based on our extensive study, geneCube data with a total of four replications for each gene by sample combination can detect a 1.3 fold gene expression change in a wide range of gene expression levels. Customers can choose a number of replications for their experiments according to accuracy requirements.

1 Experimental design including determining number of replications in terms of geneCards and hybridizations and positive and negative controls according to customer’s specific needs geneCube is infinitely flexible for experimental design
2 Design unique probe of oligo 50mers for each gene and probes have uniform hybridization condition Using Plexigen’s proprietary algorithms and software. Avoid 2nd structures and repeats. Bias toward 3’ end.
3 Coat geneCards with high quality oligo probes and QC. Form geneCubes
4 Sample preparation including RNA extraction, amplification and labeling Customer can choose to do this step
5 Hybridization
6 geneCard reading
7 Statistical data analysis for identifying gene expression patterns Plexigen will deliver customer signal intensity data and basic QC statistics if no additional statistical analysis is requested
8 Cross platform validation using RT-PCR Available for extra cost

Protein Interaction Assay Service

The geneCube™ can be used for detection of protein-protein and protein-compound interactions. For example, the geneCube can be used for parallel ELISA (Enzyme-linked immunosorbent assay) types of assays. For ELISA, an enzyme is linked chemically to a antibody or antigen. The unlabeled antigen or antibody is immobilized on a solid support, such as the bottom of a microtiter plate. For example, an antigen being immobilized on a solid support can assay a labeled antibody. The labeled antibody binds to the antigen, under the conditions where non-specific bindings are blocked. Antibody binding is detected by a reaction that coverts a colorless substrate into a colored reaction product. The ELISA type assays can be implemented on the geneCube platform in a parallel fashion. For example, 50 different antibodies can be coated on geneCards and assayed for 384 different antigens in parallel. One such cube will be equivalent to 4 X 50 = 200 microtiter plates. Besides high throughput, the geneCube platform requires a much smaller amount of sample solution for detection of 50 antigens.
