Gene Expression Experiments Using geneCube Platform Plexigen's geneCube platform is infinitely flexible for experimental design, which can meet customer's needs in gene content and accuracy requirements. A geneCube is essentially a deck of cards with each card coated with one or a number of oligo probes. The deck can be formed according to customer's gene content needs. Based on our extensive study, geneCube data with a total of four replications for each gene by sample combination can detect a 1.3 fold gene expression change in a wide range of gene expression levels. Protein Interaction Assay Service The geneCube™ can be used for detection of protein-protein and protein-compound interactions. For example, the geneCube can be used for parallel ELISA (Enzyme-linked immunosorbent assay) types of assays. For ELISA, an enzyme is linked chemically to a antibody or antigen. The unlabeled antigen or antibody is immobilized on a solid support, such as the bottom of a microtiter plate. For example, an antigen being immobilized on a solid support can assay a labeled antibody. The labeled antibody binds to the antigen, under the conditions where non-specific bindings are blocked. Antibody binding is detected by a reaction that coverts a colorless substrate into a colored reaction product. The ELISA type assays can be implemented on the geneCube platform in a parallel fashion. For example, 50 different antibodies can be coated on geneCards and assayed for 384 different antigens in parallel. One such cube will be equivalent to 4 X 50 = 200 microtiter plates. Besides high throughput, the geneCube platform requires a much smaller amount of sample solution for detection of 50 antigens.
